The Maryland General Assembly chartered the town in 1811, naming it after Williams Barnes, its earliest settler. In its formative years, Barnesville developed into a regional hub for industry and commerce, featuring two taverns, a blacksmith shop, and a resident physician named Dr. Lisle. The town's educational and religious heritage also runs deep. During the 1830s, Barnesville Academy operated on the Hays Farm under the direction of Irish immigrant Thomas Carr Lannan. St. Mary's Church was established in 1807 with the help of Jesuit Ministers from Frederick, and its churchyard contains graves of immigrant workers who died of cholera during the construction of the Chesapeake & Ohio Canal. Barnesville Baptist Church, organized in 1871, continues to worship in its original frame building on the town's east side.
Today, Barnesville remains a small but vibrant community. Incorporated in 1888, the town had a population of 144 at the 2020 census and covers 0.49 square miles. The town is governed by three commissioners elected biannually, with the commissioners selecting a president from among themselves.
