The area was first settled around 1785, with formal establishment coming in 1821 when the town was created from part of Windsor. The community of Harpursville, the principal settlement within the town, takes its name from Robert Harpur, an early developer believed responsible for assigning many classical names to communities across central New York.
The town itself was named after Nathaniel Cole, whose father settled at Cole's Hill in 1795. Located northeast of Binghamton and bordered to the north by Chenango County, Colesville spans 79.2 square miles and is traversed by the Susquehanna River flowing southward through its landscape. Interstate 88 crosses the town, providing modern connectivity alongside New York State Routes 7, 79, and 235, which link communities within and beyond the town's borders.
As of the 2020 census, Colesville had a population of 4,877 residents. The town encompasses numerous hamlets and communities, including Belden, Center Village, Doraville, Nineveh, and Sanitaria Springs, formerly known as Osborne Hollow. A notable landmark is the Harpursville United Methodist Church, listed on the National Register of Historic Places in 2006.
